染色方案
建议将以下方案用于非热启动Taq酶。
1.设置PCR反应如下:
5μL不含Mg+2的10倍的聚合酶缓冲液
2.5μL50mMMgCl2
每种5mM的2mM dNTP
2.5μL20XCyber Green
1-5单位的Taq DNA聚合酶
每种引物0.1-1μM(最终浓度)
双蒸水至终体积为50μL。
2.在热循环荧光计上进行实时PCR,并在退火或延伸步骤记录荧光信号。
注意:使用ABI序列检测系统时,请确保在WELL INSPECTOR选项卡下为被动参考选择NONE。
