实验方案
用于染色细胞的样品方案
以下程序适用于大多数细胞类型。 生长培养基,细胞密度,其他细胞类型的存在和其他因素可能影响Brdu掺入。
1)制备1-10mM DMSO储备溶液。 如果储存在-20°C,DMSO储备液可以保存6个月。
2)通过离心沉淀细胞并将细胞重悬于完全生长培养基中。 培养物中的贴壁细胞可以在盖玻片上或细胞培养孔中原位染色。
3)使用5和20μM之间的浓度添加Brdu染色并孵育60分钟过夜作为指导。 最佳孵育时间取决于细胞类型和实验目标。
4)用70-80%酒精或酸 - 乙醇固定细胞20-30分钟。
5)用PBS洗涤(3次,每次2分钟)。
6)继续进行免疫细胞化学染色技术。
