从活细胞中区分和排除死细胞可以更清晰地分离和鉴定细胞群。 Cell Meter 可固定活性染料是一大类细胞不可渗透的荧光活性染料,经过优化以匹配普通流式细胞仪的主要激发激光,例如 350、405、488、633 和 647 nm。这些染料不能渗透活细胞,但可以渗透膜受损的细胞。它们不可逆地与含胺和硫醇的蛋白质和其他细胞成分发生反应。由于膜受损的死细胞或固定细胞更容易与 Cell Meter 可固定细胞染料发生反应,因此比膜完整的活细胞染色更亮,因此这些染料可用于评估哺乳动物细胞的活细胞和死细胞状态。Cell Meter RX830 可固定细胞染料经过优化,可在634 nm激发  的深红色激光下激发,发射波长为 780/50 nm。与其他商业上类似的活力染料相比,这种可固定的活力染料更加稳定。百萤生物是AAT Bioquest的中国代理商,为您提供优质的Cell Meter RX830 可固定活性染料。

样品实验方案

简要概述

在 HHBS 中制备样品(0.5 mL/测定)

将 Cell Meter RX830 添加到细胞悬液中

在室温下对细胞染色 20 - 60 分钟

清洗细胞

修复细胞(可选)

使用 780/50 nm 滤光片(APC/Cy7 通道)通过流式细胞仪检测样品

 

溶液制备 

1.储备溶液

所有未使用的储备溶液应分为一次性使用的等分试样,并在制备后储存在-20°C下,避免重复冻融循环。
1.1Cell Meter RX830 储备液 (500X)
将 200 µL DMSO 添加到 Cell Meter RX830 小瓶中,制成 500X 储备液。

注意:未使用的 Cell Meter RX830 储备液应分成一次性使用的等分试样并储存在 -20 °C 下。避免反复冷冻/解冻循环。

 

样品操作及分析

1.根据需要处理细胞。

2.用 HHBS 或您选择的不含叠氮化物和血清/蛋白质的缓冲液清洗细胞一次。

3.在 HHBS 或您选择的不含叠氮化物和血清/蛋白质的缓冲液中,以 5 - 10 × 10 6/mL 重悬细胞。

4.将 1 µL 500X Cell Meter™ RX830 储备液添加到 0.5 mL 细胞/检测中并充分混合。

5.在室温、5% CO2 培养箱中孵育 20 - 60 分钟,避光。 注意 最佳染色浓度和孵育时间应针对不同的细胞系通过实验确定。

6.洗涤细胞两次并用 HHBS 或您选择的缓冲液重悬细胞。

7.根据需要修复细胞(可选)。

8.使用 780/50 nm 滤光片(APC/Cy7 通道)通过流式细胞仪检测样品。
