DiYO™-1 在化学上与 YOYO®-1 等效(YOYO® 是 Invitrogen 的商标)。 DiYO™-1 是一种具有与 FITC 相似的绿色荧光的碳花青二聚体。它不具有细胞渗透性,作为核复染剂和死细胞指示剂,很容易与 Cy5 和罗丹明区分开来。它在没有核酸的情况下不发出荧光,并在与 DNA 结合后产生非常明亮的荧光信号。 DiYO™-1 可在培养细胞和石蜡切片中提供强效且选择性的核染色。同时使用细胞可渗透的 Nuclear Red™ LCS1 染料和细胞不可渗透的 DiYO®-1 进行标记可用于评估细胞活力。 DiYO™-1 和 Nuclear Red™ 的消光系数都比 DNA 结合的碘化丙啶大得多。

实验方案
样品分析方案
DiYO -1工作溶液配制
使用 20 mM Hepes 缓冲液(HH 缓冲液)或您选择的缓冲液在 Hanks 中配制所需浓度的 DiYO -1 工作溶液。
注意:在初始实验中,最好尝试几种染料浓度以确定产生所需结果的最佳浓度。高染料浓度往往会导致其他细胞结构的非特异性染色。
操作步骤
注意:以下方案可适用于大多数细胞类型。生长培养基、细胞密度、其他细胞类型和因素的存在可能会影响染色。玻璃器皿上残留的洗涤剂也可能影响许多生物体的染色。
1.根据需要培养和处理细胞。
2.去除细胞培养基并固定细胞。
3.将 DiYO -1 工作溶液(1 至 10 µM)添加到细胞(悬浮细胞或贴壁细胞)中,并将细胞染色 15 至 60 分钟。
4.除去染料工作溶液并添加 HH 缓冲液或您选择的缓冲液。
5.使用 FITC 滤光片通过荧光显微镜检测细胞染色。
